英文摘要The influences of salinity on bacterial communities in a coastal area of Yuniao River in the Muping District of Yantai City, China, were analyzed by high-throughput sequencing of in situ and simulation experiments. Proteobacteria, Bacteroidetes, and Cyanobacteria were shown to have a high relative abundance in the community compositions of bacterioplankton and attached bacteria in situ. Proteobacteria occupied the highest relative abundance among all samples, while Cyanobacteria with the second highest relative abundance was inversely proportional to salinity levels in Yuniao River. Among the bacterioplankton community, ammonia-oxidizing bacteria Nitrosomonas and halophile Candidatus Nitrotoga, that have important roles in the nitrogen cycle, were with high abundance. Among the attached bacteria community, salt-tolerant bacteria Porphyrobacter had a high relative abundance that changed according to the abundance of cyanobacteria. Similar results for dominant bacterial communities in different salinities were observed in the simulation experiment. Bacteroidetes and Cyanobacteria had high relative abundance in the simulation experiment, and salinity influenced the diversity and richness of the bacterial community. This was verified by correlation analysis and principal component plot analysis. These findings could provide information and data support for the understanding of the effects of salinity on bacterial communities in coastal areas and the application of salt-tolerant bacteria.
中文摘要针对烟台市牟平区入海河流鱼鸟河生态系统,利用原位调查和室内模拟方法研究河道不同盐度(2.5‰、5‰、12‰和20‰)条件下细菌群落结构,通过高通量测序技术探究海岸带地区盐度梯度变化对细菌群落结构的影响。结果显示,河道不同盐度条件下浮游细菌和附生细菌的优势菌隶属于变形菌门(Proteobacteria)、拟杆菌门(Bacteroidetes)和蓝藻门(Cyanobacteria)。变形菌门相对丰度最高;蓝藻门次之,且蓝藻门相对丰度随着盐度增加而降低。具有氨氧化功能的亚硝化单胞菌(Nitrosomonas)和具有氧化亚硝酸盐功能的嗜盐菌Candidatus Nitrotoga为优势浮游细菌,参与水体氮循环过程。耐盐菌产卟啉杆菌(Porphyrobacter)为优势附生细菌,在藻华暴发点下游其相对丰度变化趋势与微囊藻属相似。模拟实验也得到类似结果,不同处理组优势菌如变形菌门、蓝藻门和拟杆菌门等,与原位样品相同盐度条件下的细菌群落组成相似。相关分析和主成分分析表明盐度显著影响细菌群落多样性和丰富度。研究结果可为了解盐度对海岸带菌群结构功能的影响及耐盐功能菌的应用提供理论支持。
