The invention relates to chemical, biological and electronic information sciences and particularly relates to a potential paper chip and preparation and application thereof. The paper chip is a double-layered duct system which consists of carriers, wherein an electrode system is arranged on the lower carrier; the electrode system consists of a working electrode and a reference electrode which are adhered to a sensitive membrane or a working electrode, a reference electrode and a counter electrode which are adhered to a sensitive membrane. The potential paper chip provided by the invention has the advantages of portability, low consumption of reagent, low cost, good reproducibility and easiness in operation and can be used for satisfying field immediate detection and rapid diagnosis of family diseases and the like.
