The invention relates to surface potential detection methods, in particular to a surface potential detection method based on polyion amplification effect. layer-by-layer assembly method is employed to perform chemical modification on an electrode, and a polyanion aptamer serving as a recognition molecule is fixed on the modified electrode surface, then the electrode is placed in a to-be-detected target solution for incubation at room temperature, the recognition molecule on the electrode surface is desorbed from the electrode surface, thus causing potential change, and further acquiring the concentration of the target object. The method provided by the invention has no need of making a polymer ion selective sensitive film, and the combination reaction of the target object and the polyanion aptamer occurs on the electrode surface, and the electrode potential response is rapid. More importantly, the introduction of polyanions greatly improves the sensitivity and selectivity of the electrode.
