The invention discloses a method for improving the stability of monodisperse gold nanoparticles, and belongs to the technical field of nanoparticle preparation. Said method includes the following two steps : using sodium citrate to reduce chloroauric acid to synthesize AuNPs, using 0.1 M NaOH aqueous solution to regulate and control pH value of the obtained AuNPs, and regulating pH value to 12. The advantages of the present invention are : (1) after the pH value is regulated and controlled, the charge quantity and conformation of citrate radical are changed, so that the stability of citrate radical to AuNPs is improved, and stable monodisperse AuNPs are obtained; (2) The AuNPs obtained by the method of the present invention not only realizes the separation and concentration of nano particles without adding surfactant, directly modifies sulfhydryl molecules or ions, changes from conventional water phase to organic solvent, but also slows down the coffee ring effect, and does not introduce surfactant capable of generating background signal.
