The invention relates to the field of production and preparation of plant sterol and in particular relates to a method for extracting sterol from branches and leaves of jerusalem artichoke. The method comprises the following steps : by taking branches and leaves of jerusalem artichoke as raw materials, dipping the raw materials in a petroleum ether-ethanol system, stirring and repeatedly extracting at the temperature of 5-35 DEG C, rotationally evaporating and recycling a solvent after extracting to obtain a concentrated extract, sequentially carrying out silica-gel column chromatography, gel column chromatography and separation through preparation of high performance liquid chromatography on the extract, and respectively obtaining beta-sitosterol as shown in formula (I) and stigmasterol as shown in (II) with purity being greater than 90%. The extraction method can keep natural features of products; the solvent can be recycled and reused; by extracting at normal temperatures, the volatilization loss of the solvent is reduced and the energy is saved; and petroleum ether ethanol is small in toxicity, so that the production process is relatively safe.
