|Other Abstract|| 菊芋，是一种适宜在轻盐碱地上种植且极具开发潜力的生态经济型农作物，不仅能够改良盐碱地，提高盐碱地的利用率，其块茎还可制备菊糖。菊糖作为一种功能性食品配料在西方已得到广泛应用，具有促进肠道蠕动、维持脂类代谢平衡、控制血糖、促进矿物质吸收、抗氧化等生理功能，具有良好的应用前景。根据菊糖的生理功能，为了解决目前服用阿胶产品后，出现的肠胃不适、便秘，消化系统负担过重，药效不能得到充分发挥等问题，研究菊糖在阿胶产品中的应用技术。将制备的菊糖与阿胶相结合，实现两者生理功能相互协调补充，增加阿胶产品的功能性及经济价值。|
1、将菊芋经过清洗、打浆、浸泡、压榨（滤）提取、滤液除杂、脱盐脱色、浓缩、喷雾干燥最终获得颜色纯正的天然菊糖；研究提取温度、提取时间、液料比对制备过程的影响，在单因素实验基础上，确定三因素三水平的响应面分析方法，以提取温度、提取时间、液料比为自变量，菊糖得率为因变量，进行二次回归分析，建立多元二次响应面回归模型。最终得出菊糖提取最佳条件为：温度 74.09°C，提取时间65.35min，液料比为4 mL/g。
3、研究菊糖在阿胶产品中的应用。以菊糖与阿胶为主要功能成份，制备阿胶-菊糖咀嚼片及饮料，通过感官评价确定阿胶、菊糖配比，通过单因素和正交实验研究产品的制备方法，通过动物实验评价菊糖-阿胶产品功能。实验得出：菊糖在阿胶产品中应用，能够有效促进肠道蠕动、降低血糖。; Jerusalem artichoke as a kind of ecological and economic plant with profound development potential, is suitable for planting in light saline-alkali land. Jerusalem artichoke can not only improve the utilization rate of saline-alkali land and improve ecological environment of saline-alkali land, Jerusalem artichoke tubers can also be the resources of inulin, driving the development of salt-tolerant plant production. Inulin as a functional food ingredients has been widely used in the west. Inulin possesses various physiological functions, such as maintaining balance of lipid metabolism, controlling of blood sugar level, promoting of mineral absorption, antioxidant property and so on. Inulin research has attracted more attention as to its favorable application foreground. Asini Corii Colla is a good tonic medicine with the effect of tonifying blood and arresting bleeding and nourishing yin and moistening lung, as well as the effect of anti-fatigue, anti-tumor and immunity improvement and so on. It was regarded as blood tonic “effective medicine” by doctors of all dynasties. The range of application is limited owing to the fire toxin of Asini Corii Colla. The functionality of product and economic value can be increased prominently through the combination of inulin and Asini Corii Colla.
In this thesis, inulin is prepared with the method of belt type squeezing, which can be used to explore the new technology of inulin preparation. Desalination and decoloring processes are optimized in the process of extraction. Asini Corii Colla and inulin are used as the raw material to prepare the product. The contents and results are as follows:
The natural inulin with pure color was obtained after the following extraction processes: cleaning, pulping, ultrasonic soaking, squeezing, extracting, removal of impurities, filteration, desalination, decoloration, concentration and spray-drying. According to the results of single factor experiments, three extraction parameters including the extraction temperature, the length of extraction time and liquid-to-solid ratio were as the response value of the inulin extraction yield through Box-Behnken response surface design methodology. The results were described as follows: the extraction temperature was 74.09°C, an extraction time of 65.35min and a liquid-to-solid ratio of 4mL/g. Under the optimal conditions, the maximum inulin yield was up to 86.0%.
Four kinds of ion exchange resin with best properties were chosen from ten different series combined ion exchange resin through preliminary screening. Ion exchange resin elected combines at an order of strongly basic anion exchange resin, strongly acidic cationic exchange resin, weakly basic anion exchange resin. Taking the Jerusalem artichoke extraction after preliminary treatment through the ion exchange column in turn to get the best resin combination plan through the electrical conductivity, salinity and light transmittance experiment index determination. We obtained the best combination about desalination and decoloring: D202 Strongly basic anion exchange resin, HD-8 Strongly acidic cationic exchange resin, D315 Weakly basic anion exchange resin which can reach a high Decolorization rate at 99.76% and a ion removal rate at 93.68%.
Asini Corii Colla and inulin were used as the raw material to prepare the product. The preparation method of the product was studied through the single factor and orthogonal experiment. The dosage and function between the inulin and Asini Corii Colla was determined through the animal experiment evaluation. The results of animal experiment evaluation indicated that the blood sugar levels decreased significantly. The experiment of ink propelling rates showed that this produc can effectively promote intestinal peristalsis.
